چکیده

Radioactive microspheres were used to estimate the changes in regional myocardial blood flow occurring during acute myocardial ischemia. Carbonized 15-fj, spheres were injected into the left atrium of 28 open-chest dogs and the radioactivity of selected areas determined after sacrifice. Acute occlusion of the left circumflex coronary artery produced a significant diminution in the proportion of microspheres reaching the circumflex area. In addition, there was a disproportionate decrease in endocardial radioactivity in the ischemic area (endocardial/epicardial radioactivity ratio falling from 1.17 to 0.76, P < 0.001) but not in the nonischemic area. Both nitroglycerin (0.4 mg) and propranolol (1 mg/kg) failed to cause a significant change in the ratio of circumflex to descendens radioactivity during ischemia. They did, however, cause a significant increase in the ratio of endocardial to epicardial radioactivity in both ischemic and nonischemic areas.

Effect of losartan on NOX2 transcription following acute myocardial ischemia-reperfusion

Introduction: Nicotinamide adenine dinucleotide phosphate (NADPH) oxidase-2 (Nox2) is one of the predominant sources of ROS production during myocardial ischemia-reperfusion and can be induced by angiotensin II. The evidence suggests that pharmacological blockers of renin-angiotensin system can exert direct tissue effects independent of their ability to regulate blood pressure. The mechanism...

Increase of uncoupling protein-2 expression in the ischemic rat heart

Introduction: Reactive oxygen species (ROS) have been suggested to play an important role in the myocardial damage induced by ischemia – reperfusion. One element believed to be activated by ROS and to contribute to the reduction of ROS production, is the uncoupling protein-2 (UCP2). The objective of this investigation was to explore the effect of myocardial ischemia reperfusion on cardiac UC...

Effect of regional ischemia on the left ventricular end-systolic pressure-volume relationship of isolated canine hearts.

We studied the effects of regional ischemia on the left ventricular isovolumic end-systolic pressure-volume relationship (ESPVR) in six excised, blood-perfused canine ventricles. We created different extents of regional ischemia by ligating various branches of the coronary arteries while keeping the coronary arterial pressure constant (80 mm Hg). The extent of regional ischemia (Rm) relative to...

A novel hemoglobin-based blood substitute protects against myocardial reperfusion injury.

HBOC-201 (Biopure; Cambridge, MA) is a glutaraldehyde-polymerized bovine hemoglobin (Hb) solution that is stroma free, has lower viscosity than blood, and promotes O(2) unloading. We investigated the effects of HBOC-201 in a canine model of myocardial ischemia-reperfusion injury. Dogs were anesthetized and subjected to 90 min of regional myocardial ischemia and 270 min of reperfusion. HBOC-201 ...

Reduced myocardial reflow and increased coronary vascular resistance following prolonged myocardial ischemia in the dog.

Studies were performed to determine whether an alteration in coronary vascular resistance and a reduction in the reflow phenomenon occurred in the blood-perfused, heparinized canine heart after various periods of myocaridal ischemia. Regional myocardial blood flow was measured with radioactive microspheres. Proximal left anterior descending coronary artery blood flow was measured with a periart...
